Yes, this is a legitimate message from Comcast. The email is part of a recruiting campaign to enlist consumers into the FCC’s “Measuring Broadband America Program”. The FCC has collaborated with SamKnows with the goal of accurately measuring fixed broadband and openly sharing this information to the public in a effort to improve broadband in America.

How would regular testing of broadband speed affect a subscribers data usage? I took a look through the documentation, but apart from decribing what tests are performed, it doesn't seem to detail how often they are done or how much data is transferred.

It's legit. Documenation states it runs tests every 5 min when the network is idle and comsumes 20GB of data. The router comes from the UK via FedEx. It also collects wifi signals passively. I confirmed this with the FCC. I'm a tester.
